SNP-Based Analysis of FTO Variants and Type 2 Diabetes in the Bushehr Elderly Health Cohort

Abstract

Variants within the fat mass and obesity-associated (FTO) locus have repeatedly been linked to adiposity and glycemic traits. We provide a comprehensive, methods-oriented overview for SNP analyses of FTO in relation to type 2 diabetes (T2D), using the Bushehr Elderly Health (BEH) program as an exemplar cohort design. Key considerations include SNP selection, imputation quality, population stratification, and gene–environment interactions. Evidence suggests common FTO alleles confer modest T2D risk largely mediated by BMI, though residual direct effects may persist.
